I hope you got it out by now. But if not just remove the bolts and it will stick some because of the seal. A screw driver inside the radiator hose hole and a little up ward tug and it will pop off easy. It eather has a gasket or a seal around the t stat.

Maybe air in the system. The air usualy burps after it gets hot and the T stat opens and take coolant from the resivour. Pull the cap off when its cold, push the upper hose down bellow the level of the filler neck and watch for bubbles to come out. That will allow coolant to go to the top of the closed t stat.
Did you leave anything in a hose or a cap in the hole of the tank? I have seen people leave caps on things before, rags in intakes, and such.
Maybe water pump is bad. Just because it does not leak does not mean its still good. I have seen shafts break before on water pumps and power steering most often. Or the impeler could be broken off inside.
Do you have a serpintine belt and is it routed correctly? Did you replace the water pump and get the wrong one? (reverse flow with out serpintine or vise versa)
