Change plug wires, and run injector cleaner.

Is the knock just a sound, or can you actually feel it?

If all the cylinders are getting proper fuel and spark, and you are sure one cylinder is not firing (dead), then that cylinder probably suffered mechanical problems. Do a compression check & see if a bad cylinder shows up. The noise could be a problem with the valve gear, which could also result in a dead cylinder.
